In the present study, we further tested the hypothesis that the resting pHi and pHe are closely related in the adult rat brain. The pH of cultured rat brain astrocytes was measured using 2′,7′-bis-(2-carboxyethyl)-5(6)-carboxyfluorescein and Fura-2. In the presence of 0.6 mM extracellular HCO3- at resting pHe 7.35, resting pHi of 7.32 +/- 0.03 was maintained within a narrow range. Moreover, when resting pHe was lowered to 6.9, resting pHi immediately dropped to 6.63 +/- 0.02. Addition of 10 mM HCO3- to the low pHe condition further depressed pHi to 6.44 +/- 0.04. Acidification of cultured astrocytes by nigericin or by lowering pHe by titrating with HCl resulted in a simultaneous decrease in pHi and pHe. These results indicate that cultured astrocytes in the adult rat brain have a resting pHi of 7.25. Acidic external pHe can depress resting pHi, but it does not affect resting pHi of astrocytes.
